Assay of Cigarette Smoke Solution Components

The cigarette smoke solutions were assayed for the concentration of five compounds reported to be present in cigarette smoke and known to inhibit cilia in other organs. Spectrophotometric methods were chosen that could assay these compounds in complex smoke solutions reliably without interference from other components. Appropriate dilutions of freshly made smoke solutions were made into the assay medium for each compound, and at least 5 separately generated smoke solutions of each type were assayed. birth control pills


The cyanide content of smoke solutions was assayed by a modified barbituric acid-pyridine method. Briefly, appropriate dilutions of cigarette smoke solutions and KCN standards (20-160 |xg/ml) were made in 0.1 M acetate buffer, pH 4.0. To 1 ml of standards and unknowns in 10 ml glass tubes, 25 jjlI of 50 mM sodium hypochlorite was added, and the solutions were mixed. Within 1 min, 60 jxl of barbituric acid-pyridine reagent (6.0 g of barbituric acid dissolved in a mixture of 30 ml of pyridine and 64 ml of water, and acidified with 6 ml of concentrated HC1) was added, and the solutions were mixed. After 5 min, the absorbances were determined in a 1-cm cuvet at 580 nm. All samples were read within 30 min. The cyanide content of the smoke solutions was determined from a standard curve constructed from the standard concentrations.
