Effect of Individual Smoke Constituents on CBF

For each infundibulum, CBF was first recorded in control medium (EBSS-H); then the lowest concentration of test solution was introduced into the perfusion chamber, and beat frequency was again recorded. This procedure was continued using the next increasing concentration of test solution until all concentrations had been tested. The chamber was then flushed with EBSS-H alone (washout) to determine whether CBF would return to control values. The beat frequency of hamster oviductal cilia in control medium (EBSS-H) averages 6-7 Hz at room temperature. In comparison to CBF of controls, 1.78 mM acrolein significantly inhibited CBF {p < 0.001), and beat frequency continued to decrease during the washout phase of the experiment (Fig. 1). Acrolein concentration was measured in the six types of smoke solution and ranged from 36 to 267 (jtM (Table 1). The concentration of acrolein that inhibited CBF in vitro was 7- to 50-fold more than the concentration of acrolein in MS and SS smoke solutions. buy asthma inhaler

Figure 2 shows that 1.66 mM formaldehyde significantly inhibited CBF as compared to control readings in EBSS-H {p < 0.01). In contrast to the effect of acrolein, beat frequency partially recovered following washout, but had not returned to control values at 30 min after washout. Smoke solutions have formaldehyde concentrations ranging from 50 to 249 |xM (Table 1); these concentrations are 3-17 times lower than the concentration required to inhibit CBF in vitro. Acetaldehyde, like formaldehyde, produced a dose-dependent decrease in CBF; however, a significant decrease was not observed until 45 mM was used (not shown).
