Cigarette smoke is a complex mixture of over 4000 chemicals, and cigarette smoke solutions made in physiological media likewise contain numerous different types of chemicals that include the more water-soluble components of cigarette smoke. Cigarette smoke is toxic to cilia of other organs, including the mammalian respiratory tract, and to cilia of model systems such as Paramecium and mussel gill. Various chemicals in cigarette smoke, including acrolein, formaldehyde, acetaldehyde, phenol, and cyanide, have been identified as having high toxicity to respiratory and mussel gill cilia. There is some disagreement regarding which of the ciliotoxic compounds are most likely to be important and what role they play in vivo. The purpose of this study was to identify the ciliotoxic reagent(s] in MS and SS smoke solutions that inhibit CBF and OPR using in vitro assays. Compounds that were tested were chosen because in most previous reports they were found to be ciliotoxic in other systems. The concentration of each ciliotoxic reagent was also measured in MS and SS smoke solutions to determine which components are present in sufficiently high levels to account for the inhibition of CBF and OPR observed previously.
