Role of nitric oxide synthase types II and III in early protection against endotoxin-induced lung injury: ANIMALS AND METHODS (part 1)



Animals and pharmacological treatment: Male Sprague-Dawley rats (180 to 250 g) were maintained at constant temperature (22±1°C), with a 12 h light/dark cycle in the animal facility at Jagiellonian University Medical College. They were allowed water and standard rat chow ad libitum. Anes-thesized rats (thiopental, Biochemie GmbH, Vienna, Austria; 120 mg/kg intraperitoneally) were intubated intratracheally and ventilated with room air (Ugo Basile 7025 rodent ventilator Ugo Sachs, Freiburg, Germany; tidal volume 10 mL/kg, respiratory rate 50 breaths/min) during the experiment.Polyurethane catheters were inserted into left jugular and left femoral veins for drug administration. Twenty-four rats were divided at random into four groups (consisting of six rats each) and subjected to one of the following regimens. The LPS group was administered E coli LPS (2 mg/kg/min intravenously for 10 mins; Sigma, St Louis, Missouri, USA). The L-NNA plus LPS group was administered an intravenous injection of L-NNA (10 mg/kg; Sigma) before commencement of the infusion of LPS. The L-NNA plus SNAP plus LPS group was also treated with SNAP (10 |ig/kg/min intravenously for 30 mins). SNAP was synthesized from N-acetyl-penicyllamine (Sigma) in the laboratory as previously described . Infusion of SNAP was started 30 mins after injection of L-NNA and 15 mins before infusion of LPS. The AGD plus LPS group received intravenous injections of AGD (15 mg/kg intravenously) instead of L-NNA before commencement of infusion of LPS.
