Similarly, by using a specific polyclonal antibody against NOS-III, the immunohistochemical localization of NOS-III antigen followed the signal that was obtained by the in situ hybridization technique (Figure 7). The digoxigenine- (DIG) labelled cDNA probe was hybridized to tissue section and detected with an enzyme-link immunoassay as described in Animals and Methods. Formalin-fixed, paraffin wax-embedded lung tissue taken from a rat 30 mins after lipopolysaccharide infusion was immunostained with specific polyclonal rabbit antibodies against NOS-III by the alkaline phosphatase-anti-alkaline phosphatase method.
